Oxaliplatin belongs to the group of medicines called antineoplastics. It is used to treat cancer of the colon or rectum. Oxaliplatin is usually given along with other medicines to treat cancer. Oxaliplatin belongs to a new class of platinum agent. Oxaliplatin forms reactive platinum complexes which are believed to inhibit DNA synthesis by forming interstrand and intrastrand cross-linking of DNA molecules. Oxaliplatin is a radiation-sensitizing agent. It is cell-cycle-phase nonspecific.

Amifostine is used to help prevent or lessen some side effects caused by other medicines or radiation therapy that are used to treat cancer. The thiol metabolite of amifostine is responsible for most of the cytoprotective and radioprotective properties of amifostine. It is readily taken up by cells where it binds to and detoxifies reactive metabolites of platinum and alkylating agents as well as scavenges free radicals. Other possible effects include inhibition of apoptosis, alteration of gene expression and modification of enzyme activity. Healthy cells are preferentially protected because amifostine and metabolites are present in healthy cells at 100-fold greater concentrations than in tumour cells.
